Analyses biologiques par spectrométrie de masse

Le groupe AnabioMS propose des développements en spectrométrie de masse pour l’analyse structurale et quantitative de biomolécules (protéines, peptides, lipides et/ou métabolites), dans des matrices complexes, le plus souvent d’origine biologique (cellules, organismes, sang…). Les objectifs poursuivis sont: i) l’amélioration des performances analytiques en termes de résolution, de spécificité et de gamme dynamique de détection, de capacité de multiplexage; ii) la découverte et la validation de biomarqueurs ; iii) le transfert de méthodes de dosages en clinique ou en suivi environnemental.

Axes de recherche

  • Découverte et validation de candidats biomarqueurs protéiques pour la santé humaine (infection, vieillissement, etc.) et pour la surveillance environnementale (chez les espèces sentinelles), sur la base de nouveaux concepts logiciels et de procédures analytiques ciblées (J. Lemoine, A. Salvador).

L’augmentation de l’incidence de la résistance aux antibiotiques constitue une menace majeure en santé humaine. Établir rapidement un diagnostic dans le contexte d’une infection permet à la fois une meilleure prise en charge du patient et de limiter le risque d’émergence de micro-organismes résistants. L’équipe développe de nouvelles approches protéomiques ascendantes en spectrométrie de masse, basse et haute résolution, permettant d’identifier des micro-organismes dans des prélèvements biologiques, et de quantifier la présence éventuelle d’un ou plusieurs mécanismes de résistance aux antimicrobiens. L’objectif ultime est de transférer ces méthodes en environnement clinique et de les industrialiser (start-up Weezion).

  • Augmentation de la spécificité et simplification de l’échantillonnage de mélanges complexes par le développement d’une nouvelle stratégie basée sur la photo-dissociation laser visible couplée à la spectrométrie de masse à haute résolution (M. Girod).

Afin d’améliorer la spécificité de détection de composés ciblés faiblement concentrés, nous avons développé un montage expérimental novateur, couplant la spectrométrie de masse et la photodissociation laser (LID) dans le visible afin d’ajouter une spécificité optique à la sélectivité en masse. Comme les peptides n’absorbent pas naturellement dans le domaine du visible, cette nouvelle approche s’appuie sur la dérivatisation spécifique des composés ciblés avec un chromophore pour leur conférer une réactivité de photodissociation laser à 473 nm. Seuls les peptides dérivés seront spécifiquement fragmentés en LID, améliorant les performances de dosage de protéines oxydées dans des échantillons biologiques. Nous avons en particulier appliqué cette stratégie pour l’analyse des oxydations des cystéines et la carbonylation des protéines dans un contexte de stress oxydant.

  • Développement d’approches multi-omiques intégrées en Environnement et en Santé et applications en imagerie MS (S. Ayciriex, A. Salvador).

Les approches multi-omiques ont pour objectif l’exploration simultanée de plusieurs « omiques », tels que la génomique, la transcriptomique, la protéomique, la métabolomique et bien d’autres. Ces approches sont conçues pour offrir une vision holistique permettant de mieux comprendre les interactions complexes qui règnent entre les différents niveaux d’information moléculaire au sein d’un système biologique. Dans ce contexte, notre intérêt s’est porté sur le développement de méthodes analytiques innovantes pour les approches multi-omiques (protéomique, lipidomique et métabolomique) allant de l’optimisation de la préparation des échantillons, de l’acquisition de la donnée « omique » en spectrométrie de masse haute résolution ou ciblée (approche DIA, Scout-MRM), des analyses statistiques multivariées avec l’intégration des données par des approches de machine learning (en collaboration avec la plateforme Chemod ISA). Ces développements permettent ainsi d’obtenir des informations précieuses sur les mécanismes biologiques, les maladies, les réponses à des contaminants en santé environnementale et en santé humaine.

De plus l’apport des techniques d’imagerie par MS (MALDI, ToF-SIMS) est extrêmement bénéfique dans le contexte des approches multi-omiques, en fournissant d’une part la localisation spatiale des molécules, et permettant d’autre part d’identifier de nouveaux composés jamais caractérisés chez un organisme sentinelle par exemple.
