Chromatography & Hyphenated Techniques

The group research is focused on improving multidimensional chromatographic techniques with a view to surpassing the performance of liquid chromatographic separations. Study topics are consequently dictated by the limits to be surpassed: non-targeted analyses of complex mixtures (> 500 molecules), resolution of isobars, isomers or structurally close molecules for which LC-MS/MS approaches are poorly resolving…

Two-dimensional LCxLC chromatography is a fast-growing technique, offering very high peak capacities and orthogonalities for ionizable compounds. However, methodological developments are still required for “delicate” couplings such as HILIC and RPLC for polar compounds (including sugars). Finally, neutral compounds that diagonalize in LCxLC may require the implementation of original methods such as parallel gradients, for which an understanding of the elution behavior of the molecules remains to be elucidated.

The group is also looking to innovate, by proposing research work on LCxSFC on-line coupling. SFC (supercritical fluid chromatography) generates interactions that are very different from those obtained in LC, and the combination of the two techniques presents a definite resolutive interest, recognized by the scientific community. As this coupling does not exist commercially, all facets remain to be explored, from instrumental developments (prototype, new interfaces, developments in liaison with instrumentation companies), the study of orthogonality (selectivity of columns and reasoned choice of combinations) and highlighting the potential for resolving isomers by LCxSFC-MS/MS coupling (carried out off-line) to applications for non-targeted analysis of complex matrices. Recently, the team created an online LCxSFC prototype a nd showed its high performances over a large chemical diversity (non polar and polar samples).

Finally, all new developments and proposed methodologies must be tested on real, complex samples of interest. We have identified two main areas of application, namely the improvement of industrial processes (and in particular the impact of using biosourced and/or recycled materials) and the characterization of biomass and organic waste with a view to their recovery.
